I have had a 2008 I30 Comfort for just over a year with no problem or faults until this. Unlocked the car using central locking and all the doors unlock except the rear left passenger door. I have open the central locking mechanism manually (feels a bit limp) but door still wont open from inside or either.

As the locking knob doesnt go up or down which makes me think its a combination of a packed up lock (which was the original problem i think) and the screw gradually un-lossening itself to now make it jammed You might have to sacrifice the door trim, you will need to tear/break it away to get to the lock mechanism.
